Yes... they are random generated... but you can upload yours to others players play it.

After the Chalices are generated it is static for everybody... even the drops... so you can share your chalice's Glyphs (passcode) to others players play it.

hmm then thats an option lol  i tried one of the glyphs but said i didnt meet requirements.. 

I have my second game going,  already level 35, got the blades of mercy and have kept alfred alive.  im trying to think which route might be faster
